James was the eldest son of James, 4th Duke of Hamilton and Elizabeth Gerard. He vacillated between the Jacobite and Hanoverian causes, being said to have been made Knight of the Thistle in 1722 and Knight of the Order of the Garter in 1723 by the Old Pretender and Knight of the Thistle in 1726 by George III.

He married Lady Anne Cochrane, daughter of Lord Dundonald, in 1723. The Dundonald chevron and three boars' heads are seen quartered with the duke's arms on the back of his chair. Their son succeeded as 6th Duke.

He married Elizabeth Strangeways of Dorset by whom he had no issue and in 1737 Anne Spenser of Rendlesham, Suffolk. The second of their three children succeeded as 9th Duke; the 6th Duke's two sons died young.
